Genomic technology combined with optical imaging represents a powerful approach to gain new insights into molecular events that govern the melanoma progression pathway. Constructing SAGE libraries from primary and metastatic melanoma tissues and performing microarray analysis with primary melanoma and melanoma-positive lymph node specimens, Pfaff Smith et al. and McDonald et al. identified genes with known and unknown functions that hitherto have not been shown to be expressed or have a function in early and advanced-stage melanomas and/or melanoma precursor lesions. By conducting fluorescence imaging analysis of nevocytic and melanocytic specimens probed with cyanine dye-conjugated antibodies or oligonucleotides, several of the identified genes, including ATM, were found to be expressed at high-level levels in these tissues.
